How to extract Lycium barbarum polysaccharides?
Weigh the dried and crushed wolfberry fruit, degrease it with petroleum ether: acetone (1:1), and filter out the solvent. After the residue is air-dried, monosaccharides and oligosaccharides are removed with 80% ethanol. The polysaccharide is extracted from the residue after the removal of oligosaccharides in water at 90-100°C, and the extract is concentrated under reduced pressure and then precipitated with alcohol. The precipitate is washed with absolute ethanol and acetone and dried in vacuo to obtain crude polysaccharides from Lycium barbarum. The fractional purification of crude polysaccharides is to pass the above-mentioned Lycium barbarum polysaccharides through a DEAE cellulose column, eluting with different concentrations of NaCl gradient, and concentrate the eluates of different salt concentrations under reduced pressure, then desalinate by dialysis, and obtain different results after freeze-drying.
